Description With RoboCycler Gradient 40 or 96 temperature cyclers, the gradient block can be set to generate a linear temperature range of up to 14 or 22 across the 40- or 96-well block, respectively with a temperature difference of up to 2C between adjacent wells in each row. This means you can test up to 12 different annealing temperatures in a single experiment. Once the optimal temperature has been determined, simply set the gradient block to a single temperature of up to 99C for PCR cycling.
