LIBRARY CONSTRUCTION STRATEGIES
Agencourt has experienced personnel dedicated to shotgun library construction
and verification. We use unique library construction strategies to provide
more random libraries with increased genome coverage which are customized
for the specific needs of each project.
FINISHING AND ASSEMBLY
Agencourt offers high quality finishing. Our quality standards are among
the highest in the industry, with error rates well below 1 in 10,000 nucleotides
in the assembled contigs. Our finishing strategies can include custom
primer walking or transposon insertion methodologies. We use both commercial
and proprietary bioinformatics software packages for finishing and assembly
including Phred, PHRAP and GAP 4. Paired end sequencing data allows us
to resolve repeat sequences and to maximize the level of contig ordering.
Highly experienced annotation personnel have the ability to bring
the project to full completion with in-depth sequence annotation including
nucleotide and amino acid-based homology searches. Several automated annotation
options are available including Ab initio gene prediction of genomic sequences,
homology searches using BLAST algorithms, and protein domain searches
using hidden Markov models.
