Description The Yeast ProtoArray high-density functional protein microarray is the first microarray product that enables rapid elucidation of proteinprotein interactions on a proteome scale. The Yeast ProtoArray contains 4,088 S. cerevisiae open reading frames (ORFs) expressed as 5'-GST fusions, purified and spotted in duplicate on a 1 inch x 3 inch nitrocellulose-coated slide. With the Yeast ProtoArray PPI Kit, you can identify novel protein interactions in a single day, instead of the several weeks required using alternatives such as yeast two-hybrid systems. In addition, protein interactions can be easily assayed under different conditions to better understand interaction dynamics.


Using the Yeast ProtoArray PPI Kit, protein-protein interactions screening takes four simple steps:
Prepare your protein of interest using the included ProtoArray biotinylation reagents. • Probe the Yeast ProtoArray PPI microrray using the biotinylated protein of interest • Detect positive interactions using streptavidin-conjugated Alexa Fluor 647. • Dry and scan with most standard microarray readers.

The complete kit contains two ProtoArray Yeast Proteome and two Control Microarrays, ProtoArray Buffer Module A, ProtoArray Buffer Module B, Mini-Biotinylation Module kit, Biotinylation Purification Module, Biotinylation Assessment Module, 30 l (2 mg/ml) streptavidinconjugate Alexa Fluor(r) 647, and 40 l (0.5 mg/ml) biotinylated calmodulin kinase. The ProtoArray Buffer Module A contains 30 ml of 30% BSA, 400 l 1M DTT. The ProtoArray Buffer Module B contains blocking and probing buffers, 4 Hybrislips coverslips, 2 hybridization chambers. The ProtoArray Mini-Biotinylation Module Kit contains 100 g of Biotin-XX, sulfosuccinimidyl ester, sodium salt, 50 g SA, and 20 ng biotinylated BSA. The ProtoArray Biotinylation Purification Module contains the purification resin in storage buffer, 4 spin columns, and 8 collection tubes. The ProtoArray Biotinylation Assessment Module contains 20 l Streptavidin AP, and 5 ml AP substrate and buffers.
