Description The PolarScreen Androgen Receptor (AR) Competitor Assay Kits provide a sensitive and efficient method for studying and screening potential AR ligands using fluorescence polarization (FP). The kits contain rat AR ligand binding domain protein tagged with His and GST [AR-LBD (His-GST)], and a novel, selective fluorescent androgen ligand (Fluormone AL Green in the green assay kit or Fluormone AL Red in the red assay kit) in a homogenous mix-and-read assay format.To assay:AR-LBD (His-GST) is added to the Fluormone ligand to form an AR-LBD (His-GST)/Fluormone complex, resulting in a high polarization value. This complex is then added to individual test compounds in microwell plates. Competitors displace the Fluormone ligand from the AR-LBD (His-GST)/Fluormone complex, causing the Fluormone ligand to tumble rapidly during its fluorescence lifetime which will result in a low polarization value (Figure 1). Non-competitors will not displace the Fluormone ligand from the complex, so the polarization value remains high. The shift in polarization value in the presence of test compounds is used to determine the relative affinity of test compounds for AR-LBD (His-GST).

The PolarScreen Androgen Receptor Competitor Assay Kits include purified, active AR-LBD (His-GST) protein, Fluormone AL Green ligand (green assay kit) or Fluormone AL Red ligand (red assay kit), and buffer. Store components as indicated in the assay protocol (-80C, -20C, or +4C).
