Description The EZ-Tn5™ Plasmid-Based Deletion Machine is based on the simple, highefficiency Tn5 in vitro transposition system developed by Goryshin and Reznikoff, and by York et al. for generation of nested families of deletions and inversions by intramolecular transposition into DNA targets cloned in a transposon-containing plasmid vector. Although most researchers will be interested in using the system primarily for generating deletions, about half of the intramolecular transposition events are easily-distinguished inversions, which can also be very useful for some applications. For instance, these inversion products permit easy sequencing of the opposite DNA strand using a single primer.
