Circulomics Launches Short Read Eliminator Kit to Enhance Long-Read Sequencing Read Lengths

Circulomics is launching the Short Read Eliminator Kit to perform size selection purification of high molecular weight (HMW) DNA for long-read sequencing library preparation.

This kit enhances long-read sequencing read lengths by using size selective precipitation to remove short DNA. The gentle processing ensures that HMW DNA is not damaged and recovered with high efficiency. Short DNA up to 25 kb in length are progressively depleted while DNA less than 10 kb in length is nearly completely removed. Overall DNA recovery efficiency is dependent on sample quality but generally greater than 50%. The process takes approximately 1-2 hours. Many samples can be processed in parallel using only a centrifuge.

Testing with early access users on Oxford Nanopore MinION and PromethION devices has shown that read length N50 can be increased by up to 25 kb or more based on input DNA sample quality. The method has been used to achieve read length N50 approaching 60 kb on cell line and blood samples.

Initial results using the Short Read Eliminator Kit will be presented at the 2019 AGBT General Meeting. The product will be available for sale through the Circulomics Online Store ahead of AGBT.

The Short Read Eliminator Kit can be used alongside Circulomics’ Nanobind Big DNA Extraction Kits or with previously extracted DNA.

Future Short Read Eliminator Kits will feature different size selection cutoffs to accommodate a wider array of library types and applications. Nanobind Size Select Kits that incorporate the company’s nanostructured magnetic disks are also in development to perform reaction purification during library preparation.
